Animal model of peripheral nerve injury


The sciatic nerve crush is one of the most common models of peripheral nerve injury. It is instrumental for the evaluation of the neuro-regenerative potential of test compounds. While challenging, the recovery of nerve function and axonal regeneration remains the ultimate goal in pharmaceutical development for the treatment of peripheral nerve injury.
Several techniques can be used to monitor the nerve injury and its regeneration:
■ Histology: nerve morphometry
■ Electromyography (CMAP): neuromuscular function
■ Pedobarography: weight bearing distribution

    Following the complete destruction of myelinated fibers (see week 1 illustration), spontaneous regeneration processes are observed such as debris clearance, proliferation of Schwann cells, proliferation and progressive growth of axons. By week 4 for example, many well individualized myelinated axons can be observed. See quantitative measure
