ANXIETY

  • Anxiety can be defined as a response to threats that causes nervousness and fear. In extreme cases, anxiety can have impacts on daily life and can be characterized by different symptoms for example panic attacks.

    Model of Panic Anxiety

Panic anxiety is induced in healthy volunteers by the injecting of panicogenic drugs such as choleocystokinin tetrapeptide (CCK-4) or Yohimbine. This method is commonly used as a clinical test for new anti-panic / anxiolytic drugs.
The Neurofit model of panic anxiety is performed in the rat and has been validated using the Elevated Plus Maze test (EPM). In line with the clinical finding, benzodiazepines normalize the panic anxiety status of rats challenged with CCK-4 or with Yohimbine.

  • Compound testing

Anxiolytic drugs are usually tested in this model but other treatments could also be considered. Please feel free to contact us to discuss the feasibility of your study.

Entries/time spent in the open arms in the Elevated Plus Maze
Time spent in the lit box in the Light/Dark test.

CCK-4
  • Number of entries and time spent in the open arms

YOHIMBINE
  • Number of entries and time spent in the open arms

Injection of Cholecystokinin tetrapeptide (CCK-4) enhances the anxiety status of mice.
The panic effect of CCK-4 is suppressed by Diazepam treatment.
  • Light-Dark test in mice is a well accepted model for screening anxiolytic compounds. However, not many CROs offer to conduct this test due to the fact that its sensitivity is highly dependent on the baseline level of the control animals, which is in turn dependent upon careful management of the environmental testing conditions. Neurofit has offered the Light-Dark test in mice for more than 15 years and produced consistent data throughout this time. It has been implemented not only for drug screening but also for phenotyping purposes.

Moreover, Neurofit also offers a variant of the Light-Dark test in order to mimic the CCK-4 - induced panic anxiety as modelled in human healthy volunteers.
