Animal model of inflammation-induced cognitive decline
-
Alzheimer’s disease is the most common age-related neurodegenerative disorder, characterized by the progressive degeneration of neuronal populations and the simultaneous loss of memory and cognitive functions.
Translational in vivo model of inflammation-induced cognitive deficit
NEUROFIT offers a translational in vivo model of inflammation-induced cognitive deficit. Inflammation is produced in mice by a single administration of a non-septic dose of LPS.
The model has proved to be useful in demonstrating the clinical effect of drugs or compounds of different mechanisms.

Compound testing
This model offers various possibilities of compound testing such as anti-inflammatory drugs, cognitive enhancers and/or disease modifiers… As this is a non-exhaustive list, please feel free to contact us in order to discuss our possibilities of interaction.

Endpoints
☐ Percentage of continuous and spontaneous alternations in the T-Maze (cognitive performance)
-
An amelioration of the cognitive function in LPS-mice by the administration of drugs used in humans for the treatment of Alzheimer's disease :
A lower alternation score represents a lower cognitive performance.
CSA : Continuous and Spontaneous Alternation.

Memantine : Alzheimer's disease medication acting on the glutamatergic system by blocking NMDA receptors. -

Ibuprofen : Anti-inflammatory drug. People treated with Ibuprofen for inflammatory conditions are reported to have lower rates of Alzheimer's disease. -

Donepezil : (Aricept) is a centrally active small molecule used in the treatment of Alzheimer’s disease and different cognitive disorders.

Amyloid-β intoxication
The survival test exploits the natural tendency of cells to die in culture and is therefore used to assess the neuroprotective properties of your compounds.
Amyloid-β (in vivo)
Amyloid-β i.c.v injection induces learning deficits and a dysfunction of the cholinergic system.
BACE inhibitor screening assay
Functional neuronal assay that accumulates extracellular Aβ. The system is tailored to assess the efficiency of Aβ lowering drugs such as BACE inhibitors.
